肺癌细胞转化生长因子信息传递通路功能状态研究摘要:目的探讨肺癌细胞转化生长因子β(TGFβ)信息传递通路中配体、受体及受体相互作用蛋白-1(TRIP-1)的表达功能状态。方法分别采用免疫组化技术和Northern杂交方法,检测65例肺癌患者癌组织和癌旁组织中TGFβ1、β2及相应受体(TβRⅠ、Ⅱ)蛋白水平表达状态,以及32例肺癌患者癌组织和配对正常肺组织中TRIP-1mRNA表达变化。结果TGFβ1在肺癌组织中的表达阳性率(42.3%)和表达强度(0.41±0.04)明显低于TGFβ2(73.1%,2.03±0.11)和相应的癌旁组织(66.0%,1.26±0.11),且TGFβ1β2的表达状态与肺癌细胞的分化程度有关,低分化癌明显低于中-高分化癌(分别为P<0.05,P<0.005)。与正常肺组织相比,癌组织中TβRⅠ、Ⅱ表达虽无明显降低,但其中TβRⅡ表达强度明显弱于TβRⅠ,同时,TRIP-1mRNA表达明显降低(0.66±0.15vs1.88±0.31,P<0.01)。结论肺癌细胞TGFβ信息传递通路中配体,受体及受体相互作用蛋白的表达功能存在不同程度的缺陷。TRIP-1转录水平障碍可能是造成TGFβ信息通路失活的又一机理。关键词:肺肿瘤;转化生长因子β;受体,转化生长因子β;信息传递分类号:R734.2;Q257文献标识码:A文章编号:1000-7431(2000)01-0021-04STUDYONFUNCTIONALSTATUSOFSIGNALTRANSDUCTIONPATHWAYOFTRANSFORMINGGROWTHFACTORβINLUNGCANCERCELLSHUCheng-ping(CancerResearchInstitution,HunanMedicalUniversity,Changsha,Hunan410008,China)BINLiang-hua(CancerResearchInstitution,HunanMedicalUniversity,Changsha,Hunan410008,China)ZHANFeng-huang(CancerResearchInstitution,HunanMedicalUniversity,Changsha,Hunan410008,China)Abstract:ObjectiveToexplorethefunctionalstatusofligants,receptorsandTGF-receptorinteractingprotein-1(TRIP-1)intransforminggrowthfactorbeta(TGFβ)signaltransductionpathwayinlungcancercells.MethodsAnimmunohistochemicalstudywasundertakentoanalyzeTGFβ1、β2andTGFβtypesⅠandⅡreceptorproteinonthecancerousandparacanceroustissuespecimensfrom53pateintswithprimarylungcancer.ExpressionofTRIP-1mRNAwasdetectedinthecancerousandnormallungtissuesof12patientsusingNorthernblothybridization.ResultsTheexpressionofTGFβ1proteinwasmuchlowerthanTGFβ2inthecanceroustissueandlowerthanthatinparacanceroustissue(P<0.05).Meanwhile,theexpressionstatusofTGFβproteinwasrelatedtodegreeoflungcancercelldifferentiation,theproteinlevelinpoorlydifferentiatedcancerwassignificantlylowerthanthatinwellandmoderatelydifferentiatedcancer(P<0.05).TherewasnosignificantdifferenceintheexpressivelevelofTGFβtypeⅠandⅡreceptorproteinsbetweencancerousandparacanceroustissues,buttheexpressionofTRIP-1mRNA,whichisasignalingpathwayassociatedfacotr,wasmuchlowerincanceroustissuethanthatinnormallungtissue(P<0.01).ConclusionTherearedifferentdegreeofdefectsintheexpressivefunctionoftheligants、receptorsandTRIP-1orTGFβsignaltransductionpathwayinlungcancercells:thelowerexpressionofTRIP-1mightbeanothermechanismofinactivationofTGFbetasignalingpathway.Keywords:Lungneoplasms;Transforminggrowthfactorβ;Receptor,transforminggrowthfactorβ;Signaltransduction▲肺癌发病的分子机理研究表明,除癌基因的激活和抑癌基因的灭活外,生长调控失衡是肺癌发生、发展的另一重要特征[1]。转化生长因子β(transforminggrowthfactorβ,TGFβ)具有普遍的生长抑制效应,在肺癌发生过程中的生长调控作用倍受关注。实验证明,TGFβ信息传递通路中各种信号分子的表达状态直接影响其功能的发挥。本实验分别采用免疫组化和Northern杂交方法,观察了TGFβ信息传递通路中主要信号分子即TGFβ配体(TGFβ1、β2),受体(TβRⅠ、Ⅱ)以及受体相互作用蛋白-1(TGF-β-receptorinteractingprotein-1,TRIP-1)在肺癌、癌旁和正常肺组织中的表达状况,以探讨T...
