家蚕JAK-STAT信号通路调控的基因预测#刘佳宾,钱莹,薛仁宇,曹广力,贡成良**(苏州大学基础医学与生物科学学院,江苏苏州215123)51015202530354045摘要:JAK-STAT信号通路是近几年新发现的被多种细胞因子共用的信号传导通路,在生殖细胞分化与发育、细胞凋亡、免疫应答等过程中扮演着重要的角色。通常,活化的STAT以二聚体的形式与靶基因启动子上游的回文DNA序列(TTCN2-4GAA)结合,激活转录。家蚕作为鳞翅目昆虫的模式种,已有一些信号通路包括Toll通路和Imd通路已有很多研究,但是,关于JAK-STAT信号通路还知道的很少。在本研究中,通过生物信息学分析预测了JAK-STAT信号通路调控的基因。从家蚕基因组数据库中下载所有预测基因,通过扫描搜索启动子区域(1000bp)的STAT结合序列TTCN2-4GAA,结果显示:家蚕的9293个基因,编码7271个非冗余蛋白,其基因的启动子序列中,可以发现1-6个靶序列。Go注释的结果显示,这些蛋白质参与细胞组成,分子功能和生物学过程。表明JAK-STAT通路对很多基因发挥调节作用。靶基因中超过50%的基因与结合相关,约40%的基因与细胞过程,代谢过程和催化活性相关。通常认为JAK-STAT通路在免疫反应中也发挥作用,在一些与抗病毒因子、细胞免疫效应和小分子抗菌肽(包括defensin,attacin,moricin和gloverin3)等相关基因的启动子区域可以检测到1-4个STAT结合靶序列,暗示一些抗病毒因子、细胞免疫效应物和抗病毒活性相关的抗菌肽的表达可能由JAK-STAT信号通路调控。关键词:JAK-STAT通路;家蚕;Go注释;基因;免疫反应:S881.2;Q751PredictionofgenesregulatedbyJAK-STATsignalpathwayinsilkworm(BombyxMori)LIU激abin,QIANYing,XUERenyu,CAOGuangli,GONGChengliang(SchoolofBiologyBasicMedicalScience,SoochowUniversity,激angSuSuZhou215123)Abstract:JAK-STATsignalingpathwaysharedbyavarietyofcytokineswasdiscoveredinrecentyears.Itplaysanimportantroleingrowthanddevelopment,cellapoptosisandimmuneresponse.Ingeneral,activatedSTATdimerbindstoapalindromicsequence(TTCN2-4GAA)locatedattheupstreampromoterregiontoactivategenetranscription.SomesignalpathwaysincludingTollandImdinsilkwormBombyxmori,amodelofLepidopteraninsect,havebeenwellstudied.However,littleisknownregardingJAK-STATsignalpathway.Inthepresentstudy,thegenesregulatedbyJAK-STATsignalpathwaywerepredictedbybioinformaticsanalysis.1000bpofupstreampromotersequenceoftheallpredictedgenesweredownloadedfromthesilkwormgenomedatabase,andtheSTATbindingsequenceTTCN2-4GAAweresearchedbyscanningthepromotersequences,theresultsshowedthat1to6thetargetsequencescouldbefoundintheupstreampromotersequencesof9293genescoding7271non-redundantproteins.Goannotationresultsshowedthattheseproteinswereinvolvedtocellularcomponent,molecularfunctionandbiologicalprocess,suggestingJAK-STATpathwayplayanimportantroleinmanyway.Morethan50%genesrelatedtobinding,about40%genesrelatedtocellularprocess,metabolicprocessandcatalyticactivityinthetargetinggenes.ItisconsideredthatJAK-STATplayaroleinimmuneresponse.1-4STATbindingsequencescouldbedetectedinpromoterregionofsomegenesrelatedtoanti-viralfactors,cellularimmuneeffector,andsmallantimicrobialpeptideincludingdefensin,attacin,moricinandgloverin3,implyingthattheexpressionofsomeanti-viralfactors,cellularimmuneeffectorsandantimicrobialpeptidesrelatedtoantiviralactivitymightberegulatedbyJAK-STATsignalpathway.Keywords:JAK-STATpathway;Bombyxmori;Goannotation;gene;immuneresponse基金项目:高等学校博士学科点专项科研基金(编号20113201130002)、国家自然科学基金(编号31072085)、国家重点基础研究发展计划(973计划)(编号2012CB114600)作者简介:刘佳宾,天津市人,在读硕士研究生,目前主要从事家蚕信号传导研究。liu激abin9891@126通信联...
